The parents participated in an experiment that measured how they make use of the face to judge emotions. “We found that some parents who have a child with autism process face information in a subtly but clearly different way from other parents,” Adolphs said. “This is evidence for the hypothesis that the parents with the autistic child have brains that function somewhat differently as well.”

He and other researchers are currently investigating that idea through brain imaging studies. One area of interest is the amygdala, a region located on either side of the brain in the medial temporal lobe that is known to process information about facial emotions and may have abnormal volume in both autistic individuals and their nonautistic siblings.

The finding indicates that certain aspects of autism do run in families. Although such a genetic link was noted in the 1940s in the earliest descriptions of autism, “our study adds considerable specific detail to the story,” Adolphs said.

“Our data strongly suggest that genetic factors make a substantial contribution to autism, but that does not mean that all of the cause of autism is genetic. Together with many other studies, our study argues that genetic factors play a very important role in autism, while leaving open a role for other, environmental factors,” he said.

UNC and Caltech are currently working together to follow up on the finding by looking at the neural circuitry of face processing in parents of autistic individuals, using functional MRI in a National Institutes of Health-funded study.

“We hope that this research contributes towards a cure for autism, even if only indirectly,” Adolphs said. “Once we understand better how people with autism - and their relatives - process social information like information about the faces of other people they look at, we will be in a better position to teach them strategies for social interaction and will be able to explain to them how they differ from neurotypical people.”

Piven said that this approach may disaggregate the phenotype in autism and provide new targets for genetic studies. “In other words, it may lead us to finding genes that are responsible for the face-processing component in autism,” he said.

The researchers noted that an important part of the paper is that it is not claiming all people with autism - or their parents - are ‘impaired’. Instead, they said the study shows that parents who have children with autism - like the autistic subjects themselves - are different and do things differently.

Hill New Investigator Award for her research. “We do, however, need a better understanding of this abnormal rhythm and its subtle warning signals so that we can develop smarter bedside monitors.”

While the condition is more common in people with undiagnosed heart problems, those who’ve had a previous heart attack and those with coronary artery disease, it also happens to seemingly healthy people when the body is under stress and secreting adrenaline, says Dr. Schumacher, a physiological and technological nursing professor.

Her current research focuses on what effect adrenaline has on the electrical patterns in the heart.

“The autonomic nervous system controls the heart rate by signaling our body to secrete adrenaline and increase our heart rate based on what we need – the fight or flight reflex,” she says.

Researchers already know that ventricular fibrillation occurs when the heart’s electrical system malfunctions, the electrical signals that control the pumping of the heart become rapid and chaotic causing the lower chambers of the heart to quiver instead of contract. Those chambers can no longer pump blood to the rest of the body, which leads to sudden cardiac death without defibrillation – a successful emergency shock to jump start the heart back into a regular beat.

Studying those electrical signals is what will lead to better medical equipment, Dr. Schumacher says.

Traditional cardiac tests such as electrocardiograms, which record the electrical activity of the heart and identify abnormal rhythms, and echocardiograms, which use sound waves to create a moving picture of the heart, haven’t been able to pinpoint minute changes that are a precursor to ventricular fibrillation; they only provide a picture of large scale electrical activity.

But, by using voltage-sensitive fluorescent dye, injecting it into an isolated animal model and photographing the images at 1,000 frames per second, researchers have been able to see the small picture. These minute images of ventricular fibrillation have recently led to the discovery that the electrical activity during ventricular fibrillation forms distinct patterns.

“The patterns aren’t random as we previously thought,” Dr. Schumacher says. “They actually form spiral waves that often collide with each other and spin off more spiral waves.”

Better bedside monitors will be able to detect the precursors to those spiral wave patterns so that doctors and nurses will have a two-to-three minute warning and can prevent ventricular fibrillation before it happens, she says.

To find out what role adrenaline plays in the whole process, Dr. Schumacher uses various drugs to simulate autonomic nervous system imbalance in an isolated animal heart. Then she photographs fluorescent images of the electrical activity while recording the heart’s rhythm with an electrocardiogram.

“We know that autonomic imbalance and too much adrenaline can contribute to the conditions promoting ventricular fibrillation,” she says. “This research aims to find out why.”

Anthracyclines improve survival in HER2-positive breast cancer patients

Treatment with the class of chemotherapy drugs called anthracyclines improves survival in women with HER2-total boob cancer who have previously had surgery, but it may not offer any benefit for women with HER2-negative tumors, according to a study published online December 25 in the Journal of the Public Cancer League.

Randomized clinical trials have demonstrated that treating early breast cancer with anthracycline-based chemotherapy improves disease-free and overall survival rates more than non-anthracycline-based regimens. However, the studies have demonstrated that anthracyclines may slightly increase the risk of heart damage and leukemia. Given these side effects, the greatest benefit of these regimens may be in women with breast tumors that overexpress HER2—a gene that is often amplified in tumors that respond to anthracyclines.

Alessandra Gennari, M.D., Ph.D., of the National Cancer Research Institute in Genoa, Italy, and colleagues compiled data from eight randomized controlled trials that compared anthracyclines and non-anthracyclines, and also reported HER2 status. Almost 30 percent of the patients’ tumors overexpressed HER2.

Overall and among patients with HER2-positive tumors, anthracycline-based chemotherapy produced a greater reduction in the risk of relapse or death than non-anthracycline-based regimens. However, among patients with HER2-negative tumors, there was no difference in survival between the chemotherapy regimens.

“The absence…of any effect of anthracyclines observed in patients with HER2-negative disease suggests that this group of patients could be spared unnecessary toxic effects related to the use of this class of agents and raises questions as to the appropriateness of control arms in randomized clinical trials in which anthracycline-based regimens are used in unselected patient populations,� the authors write.

In an accompanying editorial, Soonmyung Paik, M.D., of the National Surgical Adjuvant Breast and Bowel Project in Pittsburgh and colleagues point out that HER2 status alone may not be enough to determine who should receive anthracyclines, given the molecular differences among different subtypes of breast cancer.

“Optimization of adjuvant chemotherapy for patients diagnosed with breast cancer will depend on defining the baseline prognosis and chemosensitivity of each subclass of breast cancer beyond those crudely defined by HER2 status alone,� the editorialists write.
